Temporal dynamics of arbuscular mycorrhizal fungi colonizing roots of representative shrub species in a semi-arid Mediterranean ecosystem

Arbuscular mycorrhizal (AM) symbiosis plays an important role in improving plant fitness and soil quality, particularly in fragile and stressed environments, as those in certain areas of Mediterranean ecosystems. AM fungal communities are usually affected by dynamic factors such as the plant community structure and composition, which in turn are imposed by seasonality. For this reason, a one-year-round time-course trial was performed by sampling the root system of two representative shrubland species (Rosmarinus officinalis and Thymus zygis) within a typical Mediterranean ecosystem from the Southeast of Spain. The 18S rDNA gene, of the AM fungal community in roots, was subjected to PCR-SSCP, sequencing, and phylogenetic analysis. Forty-three different AM fungal sequence types were found which clustered in 16 phylotypes: 14 belonged to the Glomeraceae and two to the Diversisporaceae. Surprisingly, only two of these phylotypes were related with sequences of morphologically defined species: Glomus intraradices and Glomus constrictum. Significant differences were detected for the relative abundance of some phylotypes while no effects were found for the calculated diversity indices. These results may help to design efficient mycorrhizal-based revegetation programs for this type of ecosystems.#La simbiosis micorrícica arbuscular (AM) desempeña un papel importante en la mejora de la aptitud de las plantas y la calidad del suelo, especialmente en entornos frágiles y estresados, como los de ciertas áreas de los ecosistemas mediterráneos. Las comunidades de hongos AM suelen verse afectadas por factores dinámicos como la estructura y composición de la comunidad vegetal, que a su vez están condicionadas por la estacionalidad. Por este motivo, se realizó un ensayo de un año de duración mediante el muestreo del sistema radicular de dos especies representativas de matorrales (Rosmarinus officinalis y Thymus zygis) en un ecosistema mediterráneo típico del sureste de España. El gen 18S rDNA de la comunidad de hongos AM en las raíces se sometió a PCR-SSCP, secuenciación y análisis filogenético. Se encontraron cuarenta y tres tipos de secuencias de hongos AM diferentes que se agruparon en 16 filotipos: 14 pertenecían a la familia Glomeraceae y dos a la familia Diversisporaceae. Sorprendentemente, solo dos de estos filotipos estaban relacionados con secuencias de especies definidas morfológicamente: Glomus intraradices y Glomus constrictum. Se detectaron diferencias significativas en la abundancia relativa de algunos filotipos, mientras que no se observaron efectos en los índices de diversidad calculados. Estos resultados podrían contribuir al diseño de programas eficientes de revegetación basados ​​en micorrizas para este tipo de ecosistemas.
