Evaluation of liquid residues from beer and potato processing for the production of docosahexaenoic acid (C22:6n-3, DHA) by native thraustochytrid strains

Los residuos líquidos del procesamiento de cerveza (RB) y patata (RP) se evaluaron como fuentes de carbono para la producción de ácido docosahexaenoico (C22:6n-3, DHA) por dos especies nativas de Thraustochytriidae, M12-X1 y C41, en experimentos en matraces agitados. Los resultados se compararon con los obtenidos en las fermentaciones de glucosa, maltosa, almidón soluble y etanol. Ambas cepas produjeron la mayor concentración de biomasa (2,3 g/L) en la fermentación de RB suplementada con fuentes de nitrógeno [extracto de levadura (YE) y glutamato monosódico (MSG)]. El contenido de DHA en los ácidos grasos producidos por los thraustochytrids nativos dependió de la fuente de carbono fermentada; los ácidos grasos de la biomasa cultivada en fuentes de carbono que permitieron una menor tasa de crecimiento contenían más DHA. La mayor productividad de DHA [55,1 mg/(día L)] se obtuvo en la fermentación de RB-YE-MSG por la cepa M12-X1. En este medio, la cepa M12-X1 creció a una tasa de crecimiento específica de 0,014 h⁻¹ y el contenido total de ácidos grasos en la biomasa fue del 41,3 %. La producción de DHA por la cepa M12-X1 siguió un patrón no asociado a la tasa de crecimiento y el contenido de DHA en la biomasa disminuyó significativamente después de que cesó el crecimiento.#Liquid residues from beer (RB) and potato (RP) processing were evaluated as carbon sources for the production of docosahexaenoic acid (C22:6n-3, DHA) by two native Thraustochytriidae sp., M12-X1 and C41, in shaking flask experiments. Results were compared with those obtained in the fermentations of glucose, maltose, soluble starch and ethanol. Both strains produced the highest biomass concentration (2.3 g/L) in the fermentation of RB supplemented with nitrogen sources [yeast extract (YE) and monosodium glutamate (MSG)]. DHA content in the fatty acids produced by the native thraustochytrids was dependent on the fermented carbon source; the fatty acids from biomass grown on carbon sources that permitted a lower growth rate contained more DHA. The highest DHA productivity [55.1 mg/(day L)] was obtained in the fermentation of RB-YE-MSG by M12-X1 strain. In this medium, M12-X1 strain grew at a specific growth rate of 0.014 h(-1) and total fatty acid content in the biomass was 41.3%. Production of DHA by M12-X1 strain followed a non-growth rate associated pattern and DHA content in the biomass decreased significantly after growth ceased.
