Resumen	The use of dairy slurries as organic fertilizer amendments is a common practice in agriculture as a cost-saving measure, as well as a residue management strategy. However, concerns related to the increase in antibiotic resistance in the environment under the scope of the One Health strategy are increasing. In this study, we aimed to assess resistome enrichment driven by dairy slurry application in four southern Chile dairy farms. Slurry pits, rhizospheres of Lolium perenne amended with those slurries, and bulk soils were sampled. Thirteen antibiotic-resistance genes (ARGs, tetA, tetG, tetM, tetQ, tetW, tetX, sul1, sul2, blaCTXM, blaOXA-1, blaTEM, ermB, and dfrA1) for five antibiotic classes (tetracyclines, sulfonamides, beta-lactams, macrolides, and trimethoprim-sulfamethoxazole), two related integrases (intl1 and intl2), and total bacteria (16S rRNA) abundance was measured by quantitative PCR (qPCR). Then, the abundance profiles of two enzyme-inactivated ARGs (tetX and blaTEM) were determined. The differences between the bacterial communities inhabiting the different sample types were explored with 16S rRNA metabarcoding. In general, all measured ARGs were detected in slurries. A decreasing trend in ARG copy numbers was observed with increasing soil depth, with the exception of tetX, whose abundance increased in the bulk soil at specific farms. The tetX and blaTEM communities revealed no differences in the relative abundance of variants in any of the samples. Finally, taxonomic and structural differences were found among all sample types. Thus, the enrichment of the sampled farm soil resistomes was driven by the application of the raw slurries as fertilizer.El uso de purines de ganado lechero como enmiendas de fertilizantes orgánicos es una práctica común en la agricultura como medida de ahorro de costos y estrategia de manejo de residuos. Sin embargo, la preocupación por el aumento de la resistencia a los antibióticos en el medio ambiente, en el marco de la estrategia "Una Salud", está en aumento. En este estudio, nos propusimos evaluar el enriquecimiento del resistoma inducido por la aplicación de purines de ganado lechero en cuatro granjas lecheras del sur de Chile. Se tomaron muestras de fosas de purines, rizosferas de Lolium perenne enmendadas con dichos purines y suelos a granel. Se midieron trece genes de resistencia a antibióticos (ARG, tetA, tetG, tetM, tetQ, tetW, tetX, sul1, sul2, blaCTXM, blaOXA-1, blaTEM, ermB y dfrA1) para cinco clases de antibióticos (tetraciclinas, sulfonamidas, betalactámicos, macrólidos y trimetoprima-sulfametoxazol), dos integrasas relacionadas (intl1 e intl2) y la abundancia total de bacterias (ARNr 16S) mediante PCR cuantitativa (qPCR). Posteriormente, se determinaron los perfiles de abundancia de dos ARG inactivados enzimáticamente (tetX y blaTEM). Se exploraron las diferencias entre las comunidades bacterianas que habitan los diferentes tipos de muestras mediante metabarcoding de ARNr 16S. En general, todos los ARG medidos se detectaron en los lodos. Se observó una tendencia decreciente en el número de copias de ARG con el aumento de la profundidad del suelo, con la excepción de tetX, cuya abundancia aumentó en el suelo a granel en fincas específicas. Las comunidades de tetX y blaTEM no mostraron diferencias en la abundancia relativa de variantes en ninguna de las muestras. Finalmente, se encontraron diferencias taxonómicas y estructurales entre todos los tipos de muestras. Por lo tanto, el enriquecimiento de los resistomas del suelo de las fincas muestreadas se debió a la aplicación de purines crudos como fertilizante.
