Resumen	Angiostrongylus cantonensis is a zoonotic parasitic nematode of growing global health concern, largely due to the limited efficacy of current anthelmintics such as albendazole. In this study, cubebin-a dibenzylbutyrolactole lignan-was isolated for the first time from Drimys andina (Winteraceae), a Chilean endemic plant, and evaluated for its antiparasitic activity. Chromatographic purification of fresh leaves yielded cubebin as a 3:2 epimeric mixture, with its structure confirmed by 500 MHz NMR and single-crystal X-ray diffraction. In vitro assays demonstrated potent anthelmintic activity against both first-stage (L1) and infective third-stage (L3) larvae of A. cantonensis, with EC50 values of 4.7 and 15.3 mu M, respectively, making it approximately three times more potent than albendazole against L1 and comparably effective against L3. Cubebin exhibited no cytotoxicity toward monkey (Vero) or human (HaCaT) cell lines and no toxicity in Caenorhabditis elegans, indicating a favorable safety profile. In silico ADME analysis further revealed favorable pharmacokinetic and drug-likeness properties. These results highlight cubebin as a promising lead compound for the development of novel anthelmintic therapies targeting A. cantonensis and potentially other parasitic nematodes.Angiostrongylus cantonensis es un nematodo parásito zoonótico que genera creciente preocupación para la salud mundial, debido principalmente a la limitada eficacia de los antihelmínticos actuales, como el albendazol. En este estudio, se aisló por primera vez cubebina, un lignano dibencilbutirolactol, de Drimys andina (Winteraceae), una planta endémica chilena, y se evaluó su actividad antiparasitaria. La purificación cromatográfica de hojas frescas produjo cubebina como una mezcla epimérica 3:2, cuya estructura se confirmó mediante RMN de 500 MHz y difracción de rayos X de monocristal. Los ensayos in vitro demostraron una potente actividad antihelmíntica contra las larvas de primer estadio (L1) y de tercer estadio infectivo (L3) de A. cantonensis, con valores de CE50 de 4,7 y 15,3 μM, respectivamente, lo que la hace aproximadamente tres veces más potente que el albendazol contra L1 y comparablemente eficaz contra L3. La cubebina no mostró citotoxicidad en líneas celulares de mono (Vero) ni humanas (HaCaT), ni toxicidad en Caenorhabditis elegans, lo que indica un perfil de seguridad favorable. El análisis ADME in silico reveló además propiedades farmacocinéticas y de similitud con fármacos igualmente favorables. Estos resultados destacan a la cubebina como un compuesto prometedor para el desarrollo de nuevas terapias antihelmínticas dirigidas a A. cantonensis y, potencialmente, a otros nematodos parásitos.
